The goal of this system is to provide
an automated method to prioritize crystallized proteins
for data acquisition at the Joint
Center for Structural Genomics Structure Determination
Core. Prioritization is based on criteria that reflect the
main goals of JCSG: proteins with potential for identification
of new folds, and multidomain proteins implicated in cell
signalling.
PRIORITIZATION PROCESS: The process of prioritization is
two-tiered. First, crystals are classified according to PDB coverage, Pfam coverage, presence of mutiple
domains based on Pfam, and sequence length. After the initial
sorting process, crystal priorities are adjusted based on
a number of secondary factors. The factors selected are:
a) Presence of coiled-coil segments
b) Presence of transmembrane helices
c) Presence of prokaryotic lipoprotein lipid-binding sites
d) Archeal proteins present in the Thermotoga genome through
horizontal gene transfer
e) Absence of functional assignment (Hypothetical proteins)
f) Number of paralogues.
